Born in 1780 in Cheapside, Barclay was a Brewer, landowner, and MP for Southwark, Dundalk, and West Sussex. Although brought up a Quaker, Charles volunteered to serve in the local militia when the country was threatened with invasion from French forces under Napoleon. He and his wife, Anna Maria Kett, had seven children. He was related to famed abolitionist David Barclay of Walthamstow and Youngbury.
